The lipid assembly in the primitive cell membrane in the earliest stages of evolution and its transition to the modern cell membrane are the major questions in modern cell evolution. The complexity of the transition, such as membrane selectivity and the degree of curvature of primitive membranes, has puzzled scholars. Here, we report a microfluidic approach to mimic cellular microenvironment and study the membrane morphogenesis over time in the pioneering stage of cellular evolution. The spontaneous vesicle formation in the microfluidic channels reproduced the characteristics of primitive lipid vesicles in primitive cells in the context of a low osmolarity, high polymer concentration (Pi) cell culture environment and in a high Pi concentration liquid culture media. The vesicle morphology and the related mechanisms were characterized with fluorescent probes and atomic force microscopy (AFM). The formation of vesicles in the Pi-rich environment is attributed to the surface-active agent molecules in the medium. The formation time of vesicles in microfluidics was between 8 and 36 hours, which is much shorter than in a simulated cellular environment. This microfluidic approach may be useful to study the lipid segregation and vesicle formation and their transition to the cell membrane.1.
